Summary

The D. melanogaster 7TM receptor Frizzled, which is involved in epithelial cell planar polarity, is a unique member of the 7TM receptor superfamily because G protein-coupling is not its main signalling mechanism. The ectodomain of Frizzled has a cysteine-rich domain that interacts with its ligand Wingless (Wnt). The intracellular action of Frizzled is remarkably conserved. Ligand activation of the Frizzled receptor culminates in the production of beta-catenin that, in turn, modulates gene transcription. In D. melanogasterr, as well as frogs and mice, signal transduction by Frizzled receptors requires the participation of a low-density-lipoprotein (LDL)-receptor-related protein (LRP) for Wnt-induced signalling to Disheveled, the intracellular target of Frizzled.
